DWS offers a wide range of hypoxic workstations, from small footprint personal workstations for those with space constraints, to extremely large workstations and multi-chamber units designed to house pieces of equipment.
Hypoxic workstations allow the study of cancer biology in a setting that accurately represents the oxygen deprived tumour microenvironment (TME). The O2 concentration in Whitley Hypoxystations can range from 0.1 - 20% and this can be adjusted in increments of 0.1% allowing researchers to choose the O2 concentration that best fits their needs. The tumour microenvironment consists of hypoxic and necrotic regions where the level of O2 concentration can fluctuate. HIF-1α, which is normally broken down in the presence of O2, is stabilised and well maintained in hypoxystations compared to conventional hypoxic incubators. The presence of HIF-1α upregulates vascular endothelial growth factors (VEGF) protein which promotes the formation of new vasculature to support the tumour cells with adequate levels of O2 and nutrients. The new intricate blood supply that is formed is often disrupted and leaky and can therefore change the amount of O2 received by the tumour. To mimic this, Don Whitley Scientific have developed O2 profiling software, that allows you to rapidly cycle between different O¬2 concentrations within the desired time.
As part of our hypoxic range, DWS also offers the only workstation on the market specifically designed for housing Seahorse XF Analysers: the i2 Instrument Workstation. The combination of the i2 and Seahorse XF Analyzer permits simultaneous, real-time analysis of mitochondrial respiration and glycolysis in mammalian cells under precisely controlled hypoxic conditions. The i2 maintains an internal temperature no higher than 28°C and excludes CO2. The i2 can also be attached to a Whitley Hypoxystation via a transfer tunnel. Here samples can be prepared/ manipulated in the Hypoxystation and passed through the tunnel into the i2 workstation for further sample processing using the Seahorse XF Analyser.
