"Single Cell Multiomics and Spatial Gene Expression Analysis: Addressing the True Complexities of Cancer"

Cellular heterogeneity within and across tumors has been a major obstacle in understanding and treating cancer, and the complex heterogeneity is masked if bulk tumor tissues are used for analysis. Understanding the complex interactions between malignant cells, stromal cells, and infiltrating lymphocytes requires the ability to finely distinguish each cell type, and is prerequisite to enabling personalized cancer treatments. Whether deciphering the tumor microenvironment, decoding tumor heterogeneity, or investigating tumor-immune interactions, researchers need methodological approaches that offer the scale and resolution required to address their questions of interest to discern the different cell populations using bulk cell approaches.
The advent of rapidly developing single-cell sequencing technologies as offered by 10x Genomics with the Chromium solution, which include methods for cell-by-cell multiomics characterization of gene expression, epigenetic features, CRISPR-mediated perturbations and information about cell surface protein expression, have been applied to cancer research and led to exciting new findings.
However, the dissociation of solid tissue into single cell suspension as required for single cell analysis results in the loss of the positional context of gene expression which is of importance to understanding tissue functionality and pathological changes. With 10x Genomics Visium Spatial solution, tissue sections of 6.5 mm x 6.5 mm can be analyzed for the expression of all mRNA transcripts at a resolution of 55mm removing a major hurdle and burden in immunohistochemistry namely the limitation of available dyes or markers for labelling biomarkers.
This presentation will introduce 10x Genomics Chromium and Visium Spatial solutions for multiomics single cell and spatial gene expression and show how these illuminate the detail and dynamics of the tumor, the surrounding environment, and associations with the periphery.

Hannes (Hans Peter) Arnold studied Biology, Chemistry, and Practical and Theoretical Education at the LMU in Munich (Germany). He received his PhD on Molecular Biology at the Max-Planck-Institute for Biochemistry in Martinsried (Germany). He continued to work in several positions in industry at the interface between sales and science. He worked in the last seven years as Sales Specialist and Business Development Manager with different companies in the field of Next Generation Sequencing. He joined 10x Genomics in December 2017 and is Senior Science & Technology Advisor for Germany South and Austria.
