New PyroMark™Q24 System
Quantitative methylation and mutation analysis on several contiguous sites in parallel using Pyrosequencing® technology. This new small smart affordable system gives access to Pyrosequencing technology in an easy 24 well format for the first time.
New PyroMark™Q24 System
Pyrosequencing have now launched their PyroMark™Q24 system, which uses the same reliable Pyrosequencing technology, but now in a 24 well format, giving access to this robust and proven technology to many researchers for the first time. In the past access to Pyrosequencing technology for lower throughput lab scale analysis has been cost prohibitive for many groups as the technology has only been available in large higher throughput systems. PyroMark Q24 has also been developed following the requirements according to Annex III of the European Directive for In vitro Diagnostic Medical Devices 98/79/EC, making this the first IVDD compliant system using Pyrosequencing technology.
PyroMark Q24 is launched with a complete software package for the study of CpG methylation and mutation analysis offering:
- High resolution quantitative data for individual, consecutive sites.
- Built-in quality control for bisulfite treatment in methylation assays.
- Analysis of methylation in the presence of SNPs.
- Quantification of di-, tri-, or tetra-allelic mutations.
- All assays use sequence context as built-in quality control.
Sample Preparation for the PyroMark Q24
Sample preparation is handled by PyroMark Q24 Vacuum Prep Workstation which batch processes up to 24 DNA samples in parallel, from PCR-products to single-stranded templates, in less than 15 minutes.

PyroMark Q24 Analysis Software
The analysis software works in 2 modes, CpG and AQ (Allele Quantification), both modes can be used in the analysis of a single plate, making this an extremely flexible system ideally suited to labs working with several applications.
The software provides detailed reports in both CpG mode and AQ mode that can be exported to html, excel, text, or pdf.
CpG mode
- Quantification of multiple consecutive CpG sites in sequence context with high resolution of each individual site
- SNP:s present in CpG islands are analyzed simultaneously
- Quality assessment of both individual sites and their sequence context
- Possibility to view mean methylation values and predefined methylation ranges for quick overview of results

Plate overview showing individual methylation frequencies and quality assessment for multiple, adjacent CpG sites. Above Pyrogram trace shows the raw data from well A5. Grey areas indicate variable CpG positions; yellow area indicates the built-in bisulfite treatment control.
AQ mode
- Frequency calculations of mutations in sequence context with high resolution of each individual site
- Single variable positions
- Multiple variable positions AG/TCAATG/CATG
- Di- Tri- or Tetra-allelic mutations A/C/G/TACTGA
- Quality assessment of both individual sites and their sequence context

Results from PyroMark Q24 software illustrating a K-RAS analysis of mutations in codon 12 and 13. The software automatically analyzes mutation levels and gives a quality scoring for all results, it also provides an informative plate overview, raw data (Pyrogram®), theoretical histogram, and provides report information that can be exported to html, excel, text and pdf formats.

Grant Application Announcement
World Cancer Research Fund (WCRF) International is now inviting applications for grants worldwide to fund research into the effects of food, nutrition and physical activity on cancer.
Postdoctoral positions to work on SUMO and Ubiquitin
MRC funded 4 year posts (X2)
SCILLS funded 4 year posts (X2)
BBSRC funded 2.5 year post
Informatics Data Manager 4 year post
Enthusiastic and highly motivated postdoctoral scientists are sought to work on the role of ubiquitin and SUMO in pathways contributing to disease in humans. The successful applicants will join a team of researchers tackling these problems in the laboratory of Ron Hay. This is a well resourced laboratory in the College of Life Sciences which has excellent facilities for high-resolution mass spectrometry, X-ray Crystallography, light and electron microscopy, flow cytometry and whole genome siRNA screening.
